Weight loss medication: Postscript on genes
This is a short, but important, point if you (and your medical practitioner) are considering using GLP-1 agonists like Ozempic, Wegovy or Mounjaro.
One of the mechanisms of action of the GLP-1 agonists is to encourage the secretion of insulin, the blood sugar storage hormone.
To put it very simply, both our homegrown GLP-1 and the GLP-1 agonists kickstart the following sequence of events:
But carriers of a SNP or variant on a gene called TCF7L2 may not be good candidates for using a GLP-1 agonist. For TCF7L2 the genetic SNP or variant is the presence of the “T” allele in your version of that gene.
The research tells us that the “T” allele on TCF7L2 is a strong predictor of developing type 2 diabetes and gestational diabetes. The “T” allele is associated with higher baseline total and active GLP-1 levels. So, the problem isn’t a lack of GLP-1. The problem is that in individuals with the “T” allele, the homegrown GLP-1 or a GLP-1 agonist does not enthusiastically kickstart the sequence of events described above. So putting more GLP-1 into your system may well be “the less desirable therapeutic choice” (PMC5829963).
